Copy of the genetic makeup travels in a protein suitcase
The blueprint of all living beings is stored in their genetic material. In higher organisms this is stored in the well-protected cell nucleus. "Here a kind of copier works around the clock to make copies of the information needed at the time," says first author Jan Peter Siebrasse from the Institute for Physical and Theoretical Chemistry at the University of Bonn. The copies contain the information which the cells need to produce vital enzymes or other cell building materials. These copies consist of messenger RNA which travels on random paths to the membrane of the cell nucleus and from there through the nuclear pores into the cytoplasm which fills out the cells like jello.
Stringent quality control at the pore
The working group has found out that the messenger RNA lingers briefly at the pores in the membrane of the nucleus before it is finally transported out ? presumably for a final "quality control" or simply because it has to adjust in order to leave via the pore exit. The export process lasts in total only a few hundredths of a second to several seconds. "In all likelihood, the process needs much longer for larger, voluminous messenger RNA molecules than for smaller ones," adds Prof. Ulrich Kubitscheck, head of the working group Biophysical Chemistry and senior author of the publication.
Interestingly enough, only about every fourth collision between arriving messenger RNA and the cell nucleus leads to a successful export. Here, two kinds of processes can be distinguished: On the one hand, brief collisions with the nuclear membrane where presumably no pore is hit, and, on the other hand, those transports that are slowly aborted perhaps on account of a deficient quality control.

The RNA is packed in a type of "suitcase" made of proteins for transporting. "And it is quite a chunk," grins Prof. Kubitscheck. This is why some of his colleagues presume there are helpers on the outside of the cell's nucleus which pull the "suitcase" through the pores, a theory which the professional physicist together with the molecular biologist Jan Peter Siebrasse are currently investigating
Just what exactly happens en route from the copier to the pores has been clarified in recent years among others by Prof. Kubitscheck's working group at the University of Bonn. "Key experiments on this were undertaken by the biologist Dr. Roman Veith, whose doctorate thesis was awarded this year's Dr. Edmund ter Meer Ph.D. thesis prize from the university society," reports Prof. Kubitscheck. For these experiments the messenger RNA was altered so that it glowed when illuminated with a laser beam. This enabled the researchers to trace the path of individual molecules containing copies of the genetic material in living buccal gland cells of a mosquito type with up to 500 pictures per second. A light microscope with a high speed camera made the observation possible.

Once the transport processes between the "copier" and the cell nuclear membrane were understood, Prof. Kubitscheck and his colleagues turned their attention in recent years to the direct transport process through the nuclear pores. In order to observe this process, they took a number of years to construct a highly sensitive light microscope which works on the basis of target illumination. It creates delicate pictures of living samples and, in the process of taking pictures with high frequency, creates an unusually strong contrast.
Process is of fundamental biological interest
The question of how the messenger RNA enters the cell from the cell nucleus is of fundamental interest in biology, a fact Prof. Thoru Pederson (University of Massachusetts Medical School) underscores in his comment which accompanies the article paper presented by the Bonn-based scientists. In recent years, there have been two publications on this performed by working groups in the USA and Israel. In these studies, however, the messenger RNA has been altered with additives making the molecules at least double their volume. By contrast, the Bonn-based working group modified the messenger RNA in a negligible way, as Prof. Pederson determined.

The Philadelphia 76ers defeated the Boston Celtics, 82-75, Wednesday night at?Philadelphia's Wells Fargo Arena, setting the stage for?a seventh and series-deciding game in Boston on Saturday.

It was certainly ugly all around, but the 76ers came away with the victory, 82-75, in Philadelphia, forcing a seventh game at Boston Garden on Saturday night. Allen Iverson, the former 76ers star guard who appeared to thunderous applause before the game, might have said that the best way to win a playoff series at Wells Fargo Arena is ?practice, practice, practice.? The Celtics should have taken some of that advice to heart, because for most of Game 6, they looked completely flummoxed.

It wasn?t all bad news for the Celtics, who got 24 points from Paul Pierce, including 13 for 13 from the free throw line. And Kevin Garnett almost single-handedly kept Boston close throughout the fourth quarter by contributing 20 more. But no one else on the Celtics reached the?ten-point plateau, with both Rajon Rondo and Ray Allen tossing in nine apiece.
One of the most telling statistics for Boston was how it got schooled in the paint. ?Philadelphia handily outscored them on the inside, 42-16. One reason for that, 76ers head coach Doug Collins suggested afterward, was that he wanted to keep his team in ?attack mode? and not try to get into a shooting contest with the Celtics. And the mediocrity of this statistic is compounded when considering Boston outrebounded Philadelphia, 48-37 (including 14 offensive boards), but just couldn?t convert on second-chance points.
Rajon Rondo couldn?t set up the offense, which accounts for his unusually low six assists. And Boston didn?t help its offensive cause by turning the ball over 17 times. ?
The 76ers, though they only hit one of nine shots from three-point range, nonetheless distributed the ball well, getting five players in double-figures. The Celtics put no pressure on them in that department either, hitting only 3 of 14 from beyond the arc. At one point, second-shift guard Keyon Dooling, ordinarily a serviceable three-point shot-maker, tossed up a rainbow which found nothing but air and fell harmlessly into the floor seats behind the basket ? it perfectly summed up the Celtics? offensive trials. Boston supplemented that with a series-low 33.3% from the field ? including Rondo?s hitting only 4 of 14 field goals. This field-goal drought also spread to the bench, with the Celtics relief squad managing only five points ? all scored by Mickael Pietrus ? in just over a collective hour on the floor.
If Boston wasn?t exactly piling up the points last night, they made up for it with their personal fouls ? getting whistled 25 times to Philadelphia?s 16. This hurt particularly in the last quarter, with Ray Allen fouling out in the crucial final minutes, and 76ers guard Jrue Holiday and Andre Iguodala hitting some key free-throws as the clock waned. In fact, Holiday (who led the 76ers with 20 points) and teammate Lou Williams were seemingly making shots from everywhere Wednesday night ? particularly inside ? and that, as much as anything else, was a big source of frustration for Boston?s big men. ?
The scene now shifts to Boston, where the pressure of getting to the next round will be sitting squarely on the Celtics? shoulders. One important factor when considering the grueling nature of the playoff game schedule is that Boston will have two days rest ? which favors a skilled, yet relatively elderly and hobbled Celtics team. For them, Game 7 will have to be a statement game, with Pierce needing to get involved early and often, and Rondo piling up the assists. But eighth-seeded Philadelphia, the relatively young and inexperienced squad they are, have been remarkably poised and shown that they are capable of winning close games on the road. Regardless, Saturday will undoubtedly be another tight and hard-fought game.

Where'd the volcanic ash go? We're finding out
The huge eruption of Iceland's Eyjafjallajokull volcano in spring 2010 spewed fine aerosol particles, sulfur-rich gases and ash, causing massive air travel disruptions across Europe. Researchers studying those emissions are now shedding light on how and where that ash traveled.
"The huge economic impact of this event shows the need to describe with precision how a volcanic plume spreads through the atmosphere," Arantxa Revuelta, a researcher at the Spanish Research Center for Energy, Environment and Technology, said in a statement.
Revuelta and other scientists used satellites, laser detectors, sun photometers and other instruments to study the ash and aerosols (a term for small particles suspended in the air) as Eyjafjallajokull was erupting.
They found that different types of particles spread over different regions at different times. For example, they found very fine sulfur-rich particles over Spain and Portugal in May 2010, toward the end of the eruption. Alternatively, ash particles that reached central Europe in April were more than 20 times larger than those fine particles.

Ash, which is composed of the thicker particles, can cause severe damage to airplane engines. Fine particles, like those found over the Iberian Peninsula, are more dangerous to people on the ground because the particles are small enough to enter respiratory and circulation systems.
Together, the observations will help scientists develop and test models that predict where ash and other particles will travel after an eruption.
"During the management of the crisis it became evident that there are still no precise models that provide real-time data for delimiting an affected airspace, for example," said Carlos Toledano of the University of Valladolid, who led one of the studies.
One team is developing a model known as Fall3d, which is especially important for air travel because it predicts the concentrations of aerosol particles on the ground and in the air at different times after an eruption. Toledano and other researchers hope that fine-tuning this model will help airlines and transportation authorities make decisions in future eruptions.
The researchers' findings were published March 30 in the journal Atmospheric Chemistry and Physics, and in the March issue of the journal Atmospheric Environment.

CTIA Wireless 2012 wrap-up
Oh, how we will miss New Orleans. It's been a long and busy week in the Big Easy as tens of thousands of calories were sacrificed in the name of bringing you the most comprehensive coverage of CTIA's annual spring show. Not that we mind -- we love doing it, and it's easy justification for the dozens of beignets we downed during our stay. The event was a tad on the quiet side this year (no companies announced any buyouts, for starters) we still kept ourselves more than adequately busy sticking our noses in every nook and cranny of the show floor. Sprint confirms Direct Connect Now, will offer Push-to-Talk services via smartphone app
Sprint's been rather quiet at this year's CTIA Wireless in New Orleans, but that doesn't mean it hasn't been sitting around without making any sort of noise. This morning, the Now Network let the cat out of the bag about Direct Connect Now, which is a service that will allow any regular smartphone user the opportunity to use the company's push-to-talk services in very similar fashion to devices that come with PTT already built into the hardware. How is this made possible? Simply put, there's an app for that -- or at least, there will be one coming to Google Play at an unspecified time in the not-too-distant future. This doesn't mean Sprint is giving up on hardware-based Direct Connect devices: new models will continue to be released going forward, but more smartphone users will at least have an opportunity to take advantage of the services in one way or another. Greenland's glaciers are melting 30 faster than they were a decade ago, satellite images reveal.?
Greenland's ice sheet is on the move, with new images showing its glaciers moving 30 percent faster than they were a decade ago.
Skip to next paragraphGreenland and Antarctica are home to the two biggest blocks of?ice on Earth. As climate changes, these glacier are shrinking and the water contained in them is moving into the oceans, adding to the already rising sea level.
A glacier's velocity is a measure of how fast the ice on the surface of the sheet is flowing toward the edges of the sheet. This flow can be faster or slower, depending on how much the?glacier is melting. The faster the flow, the more water and ice mass is lost from the glacier.
"You can think of the Greenland ice sheet as a really large lake that has hundreds of those little outlet streams that are acting like conveyor belts to move ice from the middle of the ice sheet, where it's getting added by precipitation, to the edges," study researcher Twila Moon, a graduate student at the University of Washington, told LiveScience. Great glaciers
The researchers analyzed satellite images of the?Greenland glaciers?taken between 2000 and 2010. These annual images were put through a computer program to detect how quickly the ice is moving. In general, the glacial flow has sped up by 30 percent over the 10 years, Moon said.
To get a better idea of the glacier's dynamics, the researchers looked at the area's more than 200 glaciers individually. Some of these glaciers end on land, some?drop off into the sea, and the rest gradually extend their ice sheets into the water, creating an ice shelf.
The researchers saw that the glacier's type has a big impact on how quickly it flows. Land-ending ice sheets can move 30 to 325 feet (9 to 99 meters) per year, while glaciers that terminate in ice shelves move much faster, from 1,000 to more than 5,000 feet (305 to 1,600 m) per year.
The glaciers that drop off into the sea are flowing the fastest, Moon said, up to 7 miles (11 kilometers) per year and their speeds are accelerating. "The areas where the ice sheet loses the most ice are also the areas we are seeing the biggest changes," Moon said.
Where and why
The ice flow depends on the glacier's location, and therefore the local climate. Most of the high-velocity glaciers are in the northwest and southeast of Greenland, and these are also the calving, ocean-ending glaciers. [Images of Greenland's Dramatic Landscape]
Greenland is "such a vast area that the systems of precipitation and temperature that we see in the north can be pretty different from other areas. It doesn't see the same kind of summer temperature changes," Moon said. "There are important local factors that are contributing to when and how much the?glacier velocity changes."
There's no reason to think, from the new data, that the glaciers won't continue to gain speed. The result would be an increasing amount of ice and water adding to the sea level. Scientists see some good news: This average flow is actually slower than previous estimates suggested, meaning climate and sea-level rise models need to be tweaked.
"A lot of the drive behind current Greenland ice sheet and?Antarctica?studies is to ask, 'What sea-level rise can we expect?'" Moon said. "Both of these areas hold vast amounts of ice and the potential for very large sea-level rises. We need to understand what's happening on them to see what potential scenario will be realized."
The study is to be published tomorrow (May 4) in the journal Science.

US military officials released the seized documents this week in a clear attempt to sow discord among Al Qaeda leaders. Ayman al-Zawahiri is the current Al Qaeda honcho.
Everyone?s an editor, even Al Qaeda terrorists. That?s clear in the trove of documents seized last year from Osama bin Laden?s secret compound in Pakistan and released this week by the US Military Academy at West Point?s Combating Terrorism Center.

DC Decoder?s Liz Marlantes explains.The papers shed light on the personalities of key Al Qaeda operatives, including current Al Qaeda honcho Ayman al-Zawahiri, whom US intelligence officials have long accused of being arrogant and prickly.
Selective portions of the documents were released by the US military in a clear attempt to sow discord among Al Qaeda leaders still operating around the world.
One concerned Al Qaeda member ? who defense officials say may have been Mr. bin Laden ? makes margin notes in a missive written by Mr. Zawahiri, gently pointing out that perhaps he should try sounding a bit more humble.
?I?m afraid this sentence might be inappropriate and might be perceived by many people as braggadocio and an attempt to monopolize the accomplishment,? the editor notes in Zawahiri?s meditation on the impact of the Arab Spring.
The editor suggests that Zawahiri make more use of inclusive words like ?participation? and ?contribution? ? and advises Zawahiri against sounding too preachy.
To this end, the editor points out, maybe it would be a good idea to praise the role of the common people in the Arab Spring, which Al Qaeda ultimately supported because it offered the terrorist group an opportunity to take advantage of a power vacuum. ?We should say that this change was caused by the unity of the efforts of the children of this nation,? the editor proposes.
These discoveries have delighted US military officials, who released commentaries on the trove. ?Through the [Zawahiri Arab Spring] document, one can observe Al Qaeda?s editing process (reflected in the editor?s comments highlighted in green and in a bold font). While it is not clear if bin Laden himself did the editing, whoever did so has solid grammatical foundations and prefers a more self-effacing writing style than did al-Zawahiri.?
The military officials writing the commentary take digs, too, as they point out that Zawahiri chose not to take the editor?s advice. ?The edits were not included in al-Zawahiri?s final speech which was released in a video on 4 March 2011 on jihadi forums.? The commentary emphasizes that of ?the 12 proposed corrections only one appears in al-Zawahiri?s speech.?
By insinuating that Zawahiri?s editor may have been bin Laden himself, the Pentagon furthers a psychological-operations aim of portraying Zawahiri as obstinate and removed from the people he is supposed to be leading.
The insinuation also serves another purpose: to drive a wedge between bin Laden, still widely revered in terrorist circles, and the new Al Qaeda head.

Patient British Galaxy Nexus owners have finally reached a once far away moment. OEM accessories for the device are starting to appear at retailers such as Clove Technology. One such patient Nexus owner is nhuth? from the Android Central forums. The precious Desktop Dock has just shown up in the mail, and nhuth ?was good enough to share with us a couple of photos of the item along with a quick first impression.
I preordered the Galaxy Nexus desktop dock and vehicle dock from Clove.co.uk back in February and just received my desktop dock today. All that was in the box was the dock itself and quick-start guide. There are no cables included. The Galaxy Nexus fits in smoothly with a nice, quality feel. The pins work just as I would have hoped. Out back is a micro USB port and 3.5mm audio jack. Unlike the Nexus One dock, no bluetooth is included. So, there is no way to control your music with the phone out of the dock. The connection is solely through the three-pin connection.
Sounds like a bit of a mixed bag. A quality item, but functionality limited a little to what it might have been. Either way, we're glad to see these items are starting to appear in peoples hands. If anyone else has recently taken delivery of the same dock, let us know your own thoughts in the comments below.

Headhunters is slick and spritely, a mixture of corporate skullduggery and low-life slapstick that plays like The Firm meets Blood Simple.
"Headhunters" is a well-oiled, nasty machine.
That's some good acting by Hennie, but it's also good writing by co-scribes Lars Gudmestad and Ulf Ryberg. They freshen up familiar caper and thriller tropes, adapting the bestselling prose Norwegian crime writer Jo Nesbo.
This Norwegian thriller has a winning cutthroat perversity.
At a time when many mystery thrillers fall apart in the final fifteen minutes, Headhunters maintains its integrity.
The sex, nudity and violence are nonstop, but that's what makes "Headhunters" exciting entertainment.
A brisk, violent tour of the dangerous parts of Norway.
Whatever the cinematic equivalent of a kick in the pants might be, you'll find it at Headhunters.
Headhunters works best when punishing its lead excessively, continuously, and for no apparent reason.
...a first-class thriller with tension-enhancing music and Hitchcockian-quality cinematography highlighted by a sparkling performance by Aksel Hennie.
To say too much about this taut, and often quite gory, shocking and surprising twisty mystery would spoil the freezer-burn fun.
I can't remember the last time a movie surprised me as much as "Headhunters": not just its story twists and turns, which are considerable, but its continual stripping of character veneer and overall audaciousness left me breathless.
A nasty little crime thriller from Norway that twists you into a state of tense uncertainty right up to the end.
Headhunters is ridiculous, macabre, fun - see the original before it's irrevocably ruined by it's Hollywood remake.
With the twists of a cyclone and humor that on occasion edges close to farce, it's a knife-cutting satire that's not above a bellylaugh.
Acting is first rate, with a stellar turn by Aksel Hennie, and direction by Morten Tyldum is solid. I love seeing a terrific film from a place where you least expect it.

- Keep drinks to a minimum. You?re there to network and meet new people, not to party. You don?t want to fall asleep during a boring meeting and embarrass yourself, and you certainly don?t want to be known as the drunken guy at the meeting.
- Briefly say hello to anyone you already know. Then go on and meet the ones you don?t know.
- Have a short introduction ready to introduce yourself. Ten to twelve seconds is long enough.
- Have a confident attitude but don?t be cocky; you want people to like you.
- Exchange business cards with people who can benefit you directly or indirectly.
- Have a pen (and a spare) ready so you can jot notes on the back of business cards for future reference.
- It?s best not to conduct business while networking. Set a future time, such as a lunch or an appointment.
- Keep an open ear and open mind for things you may be interested in. You are someone else?s networking target too.
- Talk to as many people as you can without cutting anyone short. You can always go back to an interesting person if there?s time left at the end.
- Don?t leave immediately when the function is over. Wait and meet the speakers and other VIPs.
- If you really like the group or organization, offer to speak at a future meeting.
- The next day, send a ?nice to meet you? note to everyone from whom you have a business card. Enclose two of your own cards with the note and hand-address the envelope.
- Follow up as promised with all the people you said you would call. Do it within a week or at least send an email.
After you leave the meeting or function, you need to decide whether this is the type of function you want to attend regularly. You want to invest the time you have available where it will do you the most good. There are many choices of meetings available, so you can always try another if the first one isn?t a good fit.
Most newspapers publish a list in the Sunday or Monday edition with times and contact information. Another source of meetings is your local or nearby city business journal. Don?t be afraid to travel to another city within 100 miles to attend a new meeting; it may be well worth it. Try to budget as much of your time as you can for attending meetings, and you should see results over time.
Don?t forget about networking outside of meetings, too. A few years back, I was sitting in the waiting room of a quick oil-change place, talking to a lady with kids about our mailing lists. Her husband was in the computer business and eventually purchased one of our computer user lists. It sounds like an unlikely place to meet a business contact, but you never know who you?ll meet and when.
You can also make contacts that you can refer to friends or associates if your business can?t use them. Don?t underestimate the power of referring contacts to someone else who can help them. You will probably get a two- or three-fold return over time. Of course, you must be sure the company you?re referring to is reputable and will do a good job for them.
Networking should be automatic, wherever you are and whatever you?re doing. A good networker will always seek out new people and introduce himself. You?ll notice at Chamber of Commerce or other business meetings that most people stand around or sit with people they know or are friends with. This doesn?t get them very far in the new-contacts game. The good networker will venture up to someone standing alone and start a conversation. The objective is to meet and get to know all the new people at any meeting before it?s over. If there?s lunch or dinner, good networkers will find a table where they know few (if any) people. By the end of the meal, they want to at least be familiar with everyone seated near them. Good networkers create an opportunity out of every encounter.
Don?t forget to network when you?re out of town and learn what?s happening elsewhere in your industry. Whether you?re on a vacation or a business trip, an hour or two of time investment can pay off in big rewards. Find some time to open the yellow pages and find other businesses like yours. Call or just go visit them, ask for the owner or manager, tell him who you are, and exchange ideas. Because you?re a non-competitor from another city, you can both open up to each other. You?re bound to find something you didn?t think of and can use as soon as you get back. Give your host some ideas that you?re using effectively in your business. Of course, invite him to stop in if he ever comes to your city. Always keep an
open-door policy for anyone coming to your business from out of town. Be a visitor and a good host?it?s a valuable learning experience.
